چکیده

Among the low molecular weight RNAs which have been shown to be localized to the nucleolus are the 4 S, U3, and 8 S RNAs. Of these, the U3 and 8 S RNAs are hydrogen bonded to nucleolar 28 S RNA. The nucleolar U3 RNA migrates as an electrophoretically homogeneous band on 8% polyacylamide gels but was separated into four bands on 16% gels. Although these bands have similar nucleotide compositions, chemical analysis of the hydroxymethyl ethylene glycol derivatives of the 3’-terminal nucleosides produced by periodate oxidation and reduction with tritiated potassium borohydride has shown that these bands contain several components. Separation on DEAR-Sephadex of the oligonucleotides obtained by TI and pancreatic RNase digestion showed that three difIerent groups of terminal oligonucleotides are present. The nucleoside peak contains 3’-terminal guanosine, SS%, and @dine, 15 %. The mononucleotide peak contains terminal adenosine, 62%, uridine, 26%, and @dine, 8%. The hexanucleotide peak contains terminal uridine, 100%. On the basis of the enzymatic digestions employed, the terminal oligonucleotides are PyG, PyC, PyGAA, PyGAU, PyGAC, PyG(N~)~NrnpNrnpU.

Characterization of nucleolar antigens of normal rat liver, regenerating rat liver, and Novikoff ascites hepatoma cells following in vitro translation of polyadenylic acid-containing messenger RNA.

Nucleolar antigens of normal rat liver, regenerating liver, and Novikoff ascites hepatoma cells were transplanted in vitro from polyadenylic acid-containing messenger RNAs isolated from the respective tissues and immunoprecipitated with specific antinucleolar antibodies and Protein A. By two-dimensional gel electrophoresis of the translation products, five antigens were detected in normal rat l...

Comparison of RNase T1 fingerprints of U1, U2, and U3 small nuclear RNA's of HeLa cells, human normal fibroblasts, and Novikoff hepatoma cells.

To determine whether there are differences between the U1, U2, and U3 small nuclear RNA's of human cancer cells (HeLa cells) and human normal fibroblasts (IMR-90 cells), and between these uridine-rich small nuclear RNA's of human and Novikoff hepatoma cells, the cells were first incubated in Eagle's medium with [32P]Pi to label these RNA's uniformly. No differences were found between the RNase ...

Liver of the Rat Rabbit Antibodies to Nucleoli of Novikoff Hepatoma and Normal

SUMMARY Antinucleolar antisera were produced in rabbits immunized with whole isolated nucleoli from normal rat liver and rat Novikoff hepatoma ascites cells. These antisera produced positive nucleolar fluorescence of varying degrees in nucleoli and nuclei of Novikoff tumor, rat liver, kidney, and Walker tumor cells. Novikoff hepatoma and liver antinucleolar antisera fixed complement when combin...

Differences in nucleolar antigens of rat liver and Novikoff hepatoma ascites cells.

Antisera to nucleoli of Novikoff hepatoma ascites and normal rat liver cells were produced in rabbits by injection of whole, isolated nucleoli. These antisera have been used to compare the nucleolar antigens that were partially fractionated by differential solubilization from nucleoli. Fourteen antigens were detected by these antisera; ten of these antigens were detected by both antisera. Oucht...
